FOXO4 DRI FAQ

What is FOXO4-DRI?

FOXO4-DRI is a synthetic peptide designed to block the interaction between the transcription factor FOXO4 and the tumor-suppressor protein p53. DRI stands for D-retro-inverso: it is built from D-amino acids in reversed order, which mimics the side-chain layout of the natural FOXO4 sequence while resisting enzymatic breakdown. It was described in 2017 by Peter de Keizer's group at Erasmus University Medical Center in Rotterdam.

What has FOXO4-DRI been studied for in research?

Published research on FOXO4-DRI has been preclinical, in cell culture and mouse models, examining cellular senescence: whether disrupting the FOXO4-p53 interaction leads senescent cells to undergo apoptosis, a research area known as senolytics. It has not been tested in published human clinical trials. Findings in cells or animals do not establish effects in people.

Is FOXO4-DRI FDA approved?

No. FOXO4-DRI has not been approved by the FDA or any other regulator, and as of October 2026 it has not entered published human clinical trials. It is a preclinical research peptide, and the material sold here is a laboratory reagent only.

FOXO4-DRI vs. FOXO4: what's the difference?

FOXO4 is a natural human protein, a forkhead box O transcription factor of about 500 amino acids that regulates genes involved in stress responses. FOXO4-DRI is a short synthetic peptide derived from part of FOXO4's sequence, built from D-amino acids in reverse order, and designed to compete with the full protein for binding to p53.

FOXO4-DRI vs. fisetin: what's the difference?

Both are studied in senolytic research, but they are different kinds of molecule. FOXO4-DRI is a synthetic D-amino-acid peptide aimed at one protein-protein interaction, FOXO4 binding to p53. Fisetin is a small plant flavonoid, found in foods such as strawberries, that is studied for several cellular targets. Fisetin is not a peptide.

What are the identifiers for FOXO4 DRI?

FOXO4 DRI: molecular weight approximately 5,300 Da. Sequence: LTLRKEPASEIAQSILEAYSQNGWANRRSGGKRPPPRRRQRRKKRG (all D-amino acids, retro-inverso). Match these identifiers against the Certificate of Analysis for the lot.

Does FOXO4 DRI come with a Certificate of Analysis?

Yes. Each lot has its own Certificate of Analysis, linked on this page for the current lot of each size: 10mg (99.16% HPLC purity). Past certificates are archived in the COA library; contact us with the lot number on the vial label if a certificate is missing.

What purity is FOXO4 DRI?

99.16% (HPLC, current lot). Purity is measured by high-performance liquid chromatography (HPLC) and reported on the lot's Certificate of Analysis.

What sizes of FOXO4 DRI are available?

FOXO4 DRI is supplied as lyophilized powder in a sealed vial: 10mg. In stock and dispatched in 1–2 business days. Quantity pricing applies automatically at 5+ and 10+ vials of the same size.

How should FOXO4 DRI be stored?

Store sealed at −20 °C, dry and protected from light.
