Sermorelin Acetate FAQ

What is sermorelin?

Sermorelin is GHRH(1-29)-NH2: the first 29 amino acids of human growth-hormone-releasing hormone, the shortest fragment with full activity at the GHRH receptor. It was once an FDA-approved active ingredient, but that product was withdrawn from the U.S. market in 2008 for commercial reasons. Ours is the acetate salt, supplied for research.

Tesamorelin vs. ipamorelin vs. sermorelin

Tesamorelin and sermorelin act on the growth-hormone-releasing hormone (GHRH) receptor; ipamorelin acts on a different receptor, the ghrelin receptor (GHS-R1a). Sermorelin is the 29-amino-acid active fragment of human GHRH. Tesamorelin is the full 44-amino-acid GHRH sequence with an added trans-3-hexenoyl group that stabilizes it. Ipamorelin is a much smaller synthetic pentapeptide, Aib-His-D-2-Nal-D-Phe-Lys-NH2.

Are peptides like ipamorelin, sermorelin and CJC-1295 the same as HGH?

No. HGH is the 191-amino-acid growth hormone itself. Ipamorelin, GHRP-6, hexarelin, sermorelin, CJC-1295 and tesamorelin are much smaller synthetic peptides, called growth hormone secretagogues, that act on pituitary receptors controlling the body's own release of growth hormone: the ghrelin receptor (GHS-R1a) or the GHRH receptor. None of them contains the growth hormone sequence.

Is sermorelin FDA approved?

Sermorelin was once an FDA-approved active ingredient, but the only approved product was discontinued and withdrawn from the U.S. market in 2008 for commercial reasons. No sermorelin medicine is currently marketed under an FDA approval. The sermorelin acetate sold here is a research reagent, not a medicine, and is for laboratory research use only.

What has sermorelin been studied for in research?

Sermorelin, GHRH(1-29)-NH2, has been used for decades as a tool to study the GHRH receptor and pituitary growth hormone secretion, in pituitary cell cultures and animal models. It was tested in human clinical trials in the 1980s and 1990s, including diagnostic testing of pituitary function and studies in children with growth hormone deficiency, before its approval and later market withdrawal. Findings in cells or animals do not establish effects in people.

Is sermorelin HGH?

No. Sermorelin is a 29-amino-acid peptide, the active N-terminal fragment of growth-hormone-releasing hormone (GHRH). HGH is growth hormone itself, a 191-amino-acid protein made by the pituitary. Sermorelin acts on the GHRH receptor, upstream of growth hormone, and contains none of the growth hormone sequence.

Is CJC-1295 the same as sermorelin?

Closely related but not the same. CJC-1295 without DAC (modified GRF 1-29) is the sermorelin sequence, GHRH(1-29), with four amino-acid substitutions: D-alanine at position 2, glutamine at 8, alanine at 15 and leucine at 27. These make it more resistant to enzymatic breakdown. CJC-1295 DAC adds a lysine and a maleimidopropionyl linker that bonds to albumin. All three act on the GHRH receptor.

CJC-1295/ipamorelin vs. sermorelin vs. tesamorelin

Sermorelin, CJC-1295 and tesamorelin are all GHRH analogues acting on the GHRH receptor: sermorelin is the native 29-amino-acid fragment, CJC-1295 (no DAC) is that fragment with four stabilizing substitutions, and tesamorelin is the full 44-residue sequence with an N-terminal hexenoyl group. The CJC-1295/ipamorelin blend adds a second compound, ipamorelin, which acts on a different receptor, the ghrelin receptor (GHS-R1a).

What does "acetate" mean in sermorelin acetate, GHRP-6 acetate or hexarelin acetate?

It names the counter-ion. Peptides with basic groups, such as lysine, arginine, histidine and the N-terminus, carry positive charges that are balanced by negative ions. After purification, many peptides are converted to the acetate salt, which is preferred in research over the trifluoroacetate left by HPLC. The counter-ion adds weight, so the net peptide content of a vial is somewhat lower than the gross powder weight.

How do you reconstitute sermorelin in the lab?

Sermorelin acetate dissolves readily in water-based diluents. Let the sealed vial reach room temperature, add sterile diluent slowly down the inside wall, and swirl gently until clear; do not shake. The concentration is the labeled mass (for example 5 mg or 10 mg) divided by the volume of diluent added. Label the vial with the date and concentration and keep it refrigerated.

Does sermorelin need to be refrigerated, and does it expire?

Yes to both. Keep the lyophilized vial refrigerated at 2–8 °C, or frozen at −20 °C for long-term storage, and use it by the date on the label. Once reconstituted, sermorelin is much less stable, because the GHRH(1-29) sequence is prone to degradation in solution; keep it refrigerated and use it within the window stated for the lot.

Kisspeptin-10 vs. sermorelin

They act on different hormone systems. Kisspeptin-10 is a decapeptide (YNWNSFGLRF-NH2) that activates the kisspeptin receptor (KISS1R, also called GPR54), studied in research on the reproductive axis and gonadotropin-releasing hormone. Sermorelin is a 29-amino-acid GHRH fragment that acts on the GHRH receptor in the growth hormone axis. They share no sequence.

Is sermorelin legal?

In the United States, sermorelin is not a controlled substance and can be lawfully purchased as a research chemical for laboratory use. It is not currently marketed as an approved medicine, and selling it for human use or with health claims is unlawful. The World Anti-Doping Agency lists GHRH and its analogues, naming sermorelin, in class S2, prohibited in sport at all times. This is general information, not legal advice.

BPC-157 or TB-500 vs. CJC-1295, sermorelin or tesamorelin

These belong to unrelated research areas. BPC-157 (a 15-amino-acid gastric-protein-derived peptide) and TB-500 (derived from the actin-binding protein thymosin β4) are studied in tissue-repair models. CJC-1295, sermorelin and tesamorelin are GHRH analogues acting on the GHRH receptor in the growth hormone axis. They share no sequence, receptor or pathway.

GHRPs (GHRP-6, hexarelin) vs. GHRH analogues (CJC-1295, sermorelin, tesamorelin)

They act on different receptors. GHRP-6, GHRP-2 and hexarelin are small synthetic hexapeptides that act on the ghrelin receptor (GHS-R1a). CJC-1295, sermorelin and tesamorelin are much longer analogues of growth-hormone-releasing hormone, 29 to 44 amino acids, that act on the GHRH receptor. Both receptors are on pituitary cells and regulate growth hormone release through separate signaling pathways.

IGF-1 LR3 vs. CJC-1295, ipamorelin, sermorelin or tesamorelin

They sit at different points of the growth hormone axis. CJC-1295, sermorelin and tesamorelin (GHRH receptor) and ipamorelin, GHRP-6 and hexarelin (ghrelin receptor) are small peptides that act on pituitary receptors controlling growth hormone release. IGF-1 LR3 acts further downstream: it is an analogue of IGF-1, the hormone produced largely in response to growth hormone, and binds the IGF-1 receptor directly.
